That's just a vacuum port. You can hook your brake booster up there.

Pcv valve, does brake booster or carb go there?

Run a hose from your PCV to the 3/8" port on the back (or front, if you have an edelbrock) of your carb.

Fuel goes in pump, then out the threaded fitting into thre filter to the carb and from the second port of filter goes to return

Is this just a vent, it runs alongside return on frame rail

That may have been a tank vent line to your charcoal cannsiter...follow it back the frame and see where it goes.
